Microtubules play an important role in the transport of viral pathogens during their infection cycles. However, whether microtubule dynamics plays a role during viral pathogenesis remains to be established. Using live-cell imaging combined with gain and loss of function experiments, we demonstrate that the vaccinia virus protein F11L inhibits RhoA-mDia signalling, resulting in increased microtubule dynamics during infection. The loss of RhoA-mDia signalling also results in changes in the organization of cortical actin that allow microtubules to reach the plasma membrane. These changes in microtubule behaviour and the actin cortex are critically required for the virus to reach the plasma membrane as it exits its host.
