Toll-like receptors (TLR) mediate the innate ability of complex eukaryotes to recognize the presence of bacteria and viruses. We found that TLR5 serves as the primary means by which intestinal epithelial cells detect flagellated bacteria. While TLR5 is polarized to the basolateral surface of the intestinal epithelium and thus not normally activated by flagellated commensal bacteria, basolateral TLR5 can detect flagellin in response to a variety of invasive pathogens or colonization by pathogens with ability to modulate epithelial transport. Furthermore, upon epithelial barrier dysfunction TLR5 can be activated by flagellin released by commensal bacteria. Activation of TLR5 results in the rapid induction of genes associated with intestinal inflammation. Experiments with mice lacking TLR5 demonstrate such gene expression plays an important role in protecting the intestine from both the enteric pathogen Salmonella typhimurium and normally commensal bacteria.
