概  要:

We investigated various categories of hypermutable At-Risk Motifs (ARMs) that are poor substrates for DNA replication or repair. Defects in DNA metabolism provide other causes of genome destabilization. We highlight important biological role of strong synergistic interactions between subtle DNA metabolic defects. Such deleterious combinations of individually neutral changes severely challenge the accuracy of genome duplication. Extreme levels of genome instability can also result from interactions between subtle DNA metabolic defects and ARMs or/and environmental factors. Both concepts, ARMs and synergistic effects on genome instability proved useful in our studies of yeast DNA replication and mismatch repair.
