We examined whether M cell targeting using a protein Ag delivery system would facilitate induction of oral tolerance. We have developed a recombinant protein sigma one of reovirus genetically conjugated with ovalbumin(OVA-pσ1). Mice were fed different doses of OVA-pσ1or PBS prior to oral challenge with OVA plus cholera toxin as mucosal adjuvant. Feeding a low dose of OVA-pσ1 resulted in reduced anti-OVA antibody and CD4-positive(CD4+ ) T cell responses in both mucosal and systemic lymphoid tisseues. OVA / MHC I-Adtetramer staining revealed a significant reduction in the numbers of OVA-specific CD4+ T cells in the intestinal lamina propria but not spleen of mice fed pσ1-OVA when compared with those fed PBS. An increased frequency of CD25+, CD4+ T cells were present in the spleen of orally tolerized mice.These results show that mucosal and systemic unresponsiveness are regulated by distinct T cell subsets.
