The sculpting of blood vessels to meet the changing requirements of tissues is essential for life. Our research suggests that endothelial cell apoptosis plays an important role in this process. We have shown that transgenic inhibition of endothelial cell apoptosis disrupts vascular development and causes embryonic death in both mice and zebrafish. We found that inhibition of apoptosis also disrupts vessel development in tissue culture models of angiogenesis. We have investigated the mechanisms that control endothelial cell apoptosis. It appears that regulation of the transcriptome and of cell surface carbohydrate profiles may contribute to the apoptotic program, in addition to previously described protein-based mechanisms. Transcriptome changes may also contribute to the regulation of endothelial cell activation by inflammatory mediators and by the survival factor VEGF-A. In addition, it appears that transcriptome changes may act downstream of the well-known apoptotic regulator Bcl-2. We are particularly interested in the possibility that the co-regulation of large numbers of transcripts by a small degree may powerfully regulate the fate of these cells. We are now using transgenic embryoid bodies to map the function of individual molecules in endothelial cells. We hope that soon we will be able to use and endothelial cell gene regulatory networks to map the synergistic function of multiple molecular signals in endothelial cells. The ultimate aim of this work is to expand our knowledge of the biology of blood vessel walls, and also reveal novel targets for the therapy of cardiovascular disease.
