演題: M期染色体セントロメアにおけるSUMO修飾の役割 (Role of mitotic SUMOylation on functions & organization of mitotic centromeres)
概要:

Accurate chromosome segregation in mitosis is an essential process to maintain complete genomic information in each daughter cell. Failure of faithful chromosome segregation during anaphase causes developmental defects and contributes to tumor progression by alteration of the genome. Both genetic and biochemical studies indicate that the posttranslational protein modification by Small Ubiquitin-like MOdifier (SUMO), SUMOylation, has an essential role in normal chromosome segregation. We have approached to elucidate molecular mechanism of SUMOylation on chromosome segregation by using Xenopus egg extracts cell-free assay.
Our results determine that SUMOylated proteins are enriched at mitotic centromeres and could regulate their enzyme activity. In addition, SUMOylation of centromeric proteins can provide binding site for other cellular proteins that have SUMO-interacting motif (SIM) on their primary structure for efficient recruitment of SIM-containing proteins at mitotic centromeres. Identified these SUMO-dependent centromeric proteins, which have SIMS, includes histone modification enzymes and nucleosome remodeling factors. Suggesting mitotic centromeric SUMOylation could regulate histones on mitotic centromeres. Consistent to these findings, we found mitotic SUMOylation contribute to establishment of epigenetic information at mitotic centromeres. With our recent findings, I would like to discuss potential role of mitotic SUMOylation on regulation of centromeric functions and on organization of vertebrate centromeric structure.
